New drug strategy attacks resistant leukemia and lymphoma

Scientists at the Dana-Farber/Children's Hospital Cancer Center have developed an anti-cancer peptide that overcomes the stubborn resistance to chemotherapy and radiation often encountered in certain blood cancers when the disease recurs following initial treatment.

The strategy could pave the way for much needed new therapies to treat relapsed and refractory blood cancers, which are difficult to cure because their cells deploy strong protein "deflector shields" to neutralize the cell death signals that chemotherapy agents used against them initially, say the researchers.

The prototype compound, called a "stapled BIM BH3 peptide," is designed to disable the cancer's defenses by hitting a family of protein targets that regulate cell death.

In proof-of-concept studies in mice with transplanted, drug-resistant leukemia tumors, the compound alone suppressed cancer growth, and when paired with other drugs, showed synergistic anti-cancer activity, say researchers led by Loren Walensky, MD, PhD, of Dana-Farber/Children's Hospital Cancer Center.

Their paper has been posted online by the Journal of Clinical Investigation and will appear in the journal's June issue. Walensky is the senior author and James LaBelle, MD, PhD, is the first author.

A cell's "fate" ? when and whether it lives or dies ? depends on a tug-of-war between pro-death and anti-death forces within the cell that serve as a check-and-balance system to maintain orderly growth. The system is regulated by the BCL-2 family of proteins, which contains both pro-death and pro-survival members.

When cells are no longer needed or are damaged beyond repair, the body activates pro-death BCL-2 proteins to shut down mitochondria ? the power plants of the cell? resulting in an orchestrated cellular destruction known as apoptosis, or programmed cell death.

Many cell-killing cancer treatments work by triggering these "executioner proteins" to cause tumor cells to commit suicide in this fashion. But cancer cells can escape their death sentence ? and even become immortal ? by hyperactivating the survival arm of the family; these proteins intercept the executioner proteins and block their lethal mission.

"When cancers recur, they activate not just one type of survival protein, but many," explains Walensky, whose laboratory has extensively studied the cell-death system and makes compounds to manipulate it for research and therapeutic purposes.

"It's as if relapsed cancers 'learned' from their initial exposure to chemotherapy such that when they come back, they put up a variety of formidable barriers to apoptosis," he adds. "To reactivate cell death in refractory hematologic cancers, we need new pharmacologic strategies that broadly target these obstacles and substantially lower the apoptotic threshold."

When cancers specifically rely on one or two survival proteins, treating them with selective BCL-2 inhibitors can be very effective at eliminating the cancer cells' survival advantage. But relapsed cancers often evade such agents by deploying a battery of alternate survival proteins, so what's needed, Walensky says, are "next-generation" compounds that can block a wider range of survival proteins without jeopardizing normal tissues.

In the current research, the scientists built a chemically-reinforced peptide containing the death-activating BH3 domain of an especially potent killer protein, BIM, which is able to tightly bind with and neutralize all of the BCL-2 family survival proteins. This 'stapled' peptide, which incorporates the natural structure and properties of BIM BH3, not only disables the survival proteins, but also directly activates pro-death BCL-2 family proteins in cancer cells, making them self-destruct. Importantly, non-cancerous cells and tissues were relatively unaffected by the treatment.

"The diversity of BCL-2 family survival proteins blunts the anti-tumor activity of essentially all cancer treatments to some degree," Walensky points out. "By using Nature's solution to broad targeting of the BCL-2 pathway with a stapled BIM BH3 peptide, our goal is to eliminate cancer's protective force field and enable the arsenal of cancer treatments to do their job."

USF baseball advances to Big East tournament final

The University of South Florida baseball team will play for the Big East Conference tournament championship.

After two Saturday elimination-game victories against Connecticut ? 1-0 during the day and 2-1 at night ? the Bulls (38-21) meet St. John's (36-21) in the championship game at noon on Sunday at Bright House Field in Clearwater.

The Bulls are expected to start ace senior left-hander Andrew Barbosa, a first-team All-Big East selection.

St. John's advanced with a 2-1 victory against Notre Dame.

Today's winner will receive an automatic bid to the NCAA tournament. USF, which hasn't played at an NCAA Regional since 2002, might already be in line for an at-large bid because of the four-game win streak that has propelled the Bulls into the Big East final.

The Bulls are playing in their first conference tournament baseball final since 2001, when they played in Conference USA. USF's last conference baseball tournament championship was 1995 in the Metro Conference.

Coach Lelo Prado's Bulls came into Saturday with an 0-5 mark against UConn at the Big East tournament. But USF completed a sweep in the night game, getting six strong innings from Nick Gonzalez and scoring both runs on wild pitches.

The Bulls began on the right foot in the opening game as senior right-hander Derrick Stultz (9-1), who missed the past two seasons with injury, tossed a complete-game three-hit shutout and helped to preserve USF's staff.

It was only the second complete game of Stultz's career, along with a win against St. John's on May 11, 2008.

Stultz, who faced 34 batters, got all the support he needed on Jimmy Falla's solo home run in the top of the fourth inning.

Chesapeake Energy selling half a million acres in Colorado, Wyoming

HOUSTON?? Chesapeake Energy Corp. is selling oil and gas assets in Colorado and Wyoming as it seeks to raise revenue to fund its cash-strapped operations.

According to a prospectus listed by the transaction-advisory firm helping Chesapeake, about 503,863 net acres of land in the northern DJ Basin are being listed. The area includes both conventional and unconventional reservoirs. Last year, Chesapeake sold acreage there to China's Cnooc Ltd.

The move is part of Chesapeake's bid to raise up to $11.5 billion from the sale of assets this year to help pay down debt and run its expensive drive into oil-rich shale areas. The company recently said it would seek to divest noncore assets in addition to the widely announced sale of its oil-rich land in the Permian Basin and the listing of a joint venture in the Mississippi Lime in the central U.S.

Analysts have said they are concerned about the company's liquidity, which has been sapped by decade-low levels for natural-gas prices, by its high levels of debt and by the steep requirements of its drilling program.

From Canada Down To Argentina, The Oil Flows

    Juan Forero for NPR

Like countries throughout the Americas, Argentina is feverishly drilling for oil and gas. Workers are shown here at a derrick in the desert in southern Argentina.

As the wind whips across the scrub grass in southern Argentina, a crane unloads huge bags of artificial sand for oil workers preparing for the hydraulic fracturing, or fracking, of a well.

Water mixed with chemicals and tiny ceramic beads are then blasted underground at high pressure. This mixture helps create fissures, allowing oil and natural gas to flow.

Energy analysts believe there are billions of barrels of oil and gas buried in a desert-like patch in Patagonia.

In many places in the Americas, oil discoveries are being made and production is rising. In the U.S., for example, the same kind of technology has permitted oil companies in North Dakota to ramp up production to nearly half a million barrels a day.

The growing production in the Americas is changing the world's energy equation, with the U.S. now less dependent on Persian Gulf oil.

German Perez oversees a team of technicians from Schlumberger, the Houston-based oil services giant that's drilling in southern Argentina.

    More Oil From The Americas

Four of the world's top nine producers are in the Americas:

    1. Saudi Arabia

    2. Russia

    3. United States

    4. China

    5. Iran

    6. Canada

    7. Mexico

    8. United Arab Emirates

    9. Brazil

    Source: U.S. Energy Information Administration

Perez says the day could come when thousands of wells pockmark the land above Vaca Muerta, the huge energy-rich formation here.

"Yes, yes," he says, "I think that's what's going to happen. The projections are very positive."

    A Trend Throughout The Americas

The projections are positive up and down the Americas.

Production is rising fast in Canada, Colombia, Brazil and in the United States, which increased output by 1.7 million barrels in just six years.

RoseAnne Franco of the energy consulting firm Wood Mackenzie has been closely tracking the rise in production.

She says there's a twofold reason for the rise: high oil prices and technologies such as fracking that are unlocking the potential for hard-to-reach oil and gas.

"You are seeing that in North America, but you are also seeing that in Latin America," she says. "So areas that are more difficult to produce, such as heavy oil in Venezuela, the pre-salt in Brazil, it's now more economical."

Oil companies are also drilling in the Arctic Circle, in the heart of the Amazon, off the Falkland Islands and in the waters off French Guiana.

This new scenario comes as exports to the United States from all but one major OPEC country have fallen in recent years.

More than half of all U.S. imports now come from countries in the Americas, while just 22 percent comes from the Persian Gulf.

That doesn't mean that the oil and gas in the Americas will be easy to produce ? or that countries with large, undeveloped fields will be able to get them online fast.

    Argentine Policies Raise Concerns

In Argentina, some oil analysts consider the government's populist policies an obstacle to development.

Crowds recently celebrated on the streets after Congress unanimously approved the expropriation of the country's biggest energy firm, YPF, from its Spanish owners.

    Juan Forero for NPR

Oil workers in southern Argentina help guide a load of artificial sand that is then blasted at high pressure with waters and chemicals into a well, the process known as hydraulic fracturing, or fracking.

That has spooked other oil companies.

Their concern wasn't lost on Ruben Etcheverry. He's among the regional energy officials in Patagonia who are trying to assure oil companies that they're welcome and needed to develop potentially lucrative formations.

"What we need is more technology and investors," he says. "And I think that we have not convinced but talked with them, and they have understood that it would be for them a good activity, a good business, also to establish here."

Some major oil companies are already in Argentina, among them ExxonMobil. American companies better known for their fracking technology are here, too, such as Apache and EOG Resources.

One of the veterans of many an oil exploratory well is Juan Carlos Montiel of YPF, the Spanish firm that was recently nationalized by Argentina.

Wearing a hard hat and steel-capped boots, he walks amid roaring pumps as they rev up to drill.

He talks excitedly about the 1,200-horsepower pumps that blast water and how the operation relies on huge water containers, hose lines and computerized equipment. With this exploration that's taking place, he says, Argentina will greatly increase its production of gas and oil.

Home ? Your Pets ? Pet estate planning: It?s easier than ever to include your pets in your estate ?

    May 26, 2012 by Admin ? ?

Henry loves fetching his tennis ball and playing tug with his stuffed animals. About 6 years old, the Shih Tzu mix likely has many good years left.

In a way, that worries his owner.

Fern Edson adopted Henry from the PAWS Chicago animal shelter almost four years ago. Her age: 84. I really didnt think about the age difference, Edson said. About 18 months later, I started thinking about it.

Edson, whose investment banker husband died 12 years ago, has found peace of mind about Henrys future, however. Shes enrolled in PAWS Guardian Angel pet care program, in which donors who leave bequests to the no-kill shelter are assured it will care for animals that outlive their owners.

Its one of several ways that pet lovers are making provisions for their animals in the event they precede them in death. In recent years, it has become easier than ever to make estate-planning arrangements for pets.

All but a handful of states, for example, have passed laws that permit pet owners to pass on assets for their care. Illinois began allowing residents to create trusts for their pets in 2005. Since then, awareness of pet trusts has risen, and animal law is one of the fastest-growing legal specialties, according to the American Bar Association.

The tempo of inquiries and the number of actual executed pet trusts has increased in the last couple of years, said Michael Fitzgerald, a lawyer with St. Charles-based law firm Fitzgerald amp; Perry, which has worked on pet trusts.

If pet owners dont make arrangements, the outcome for their animals can be uncertain.

Take Angel, a 1-year-old female cat that meowed to passersby recently from her cage at the Anti-Cruelty Society of Chicago. Society spokeswoman Trisha Trinco said Angel was dropped off with two other cats April 28 after their owner died. Angel was spayed May 4 and remains at the shelter. The other cats, Peggy and Marsha, have been adopted.

Some people leave bequests for their pets in wills. But that isnt always the best option because the purpose of a will is to distribute property, and instructions are unenforceable, according to the 2010 book Petriarch: The Complete Guide to Financial and Legal Planning for a Pets Continued Care. In other words, Jane might get the house and the cat, but theres nothing in the will that compels her to keep the house maintained or the cat fed.

Wills are also subject to the probate process. A recent case in point is the Berwyn woman who died in December and stipulated in her will, written in 1988, that any animal in her care at the time of her death would be euthanized. The woman left most of her $1.4 million estate to animal causes, which suggests she cared deeply about pets. Still, her will sought a death sentence for Boots, a healthy 11-year-old cat. The wills executors at Fifth Third Bank, persuaded a Cook County probate judge last month to set aside that provision and spare the cats life. The bank then gave more than $2,000 to Chicago no-kill shelter Cats Are Purrsons Too to care for Boots while attempts are made to find her a new home.

Estate planners and lawyers say what happened in the Boots case is one reason why setting up a pet trust is better than dealing with pet issues in a will. Trusts take effect quickly in the event of the owners death or incapacitation compared with the execution of a will, a process that can drag on for weeks, if not months or years, in probate court.

Jeffrey Schmidt, Fifth Third trust officer, said if pet trusts are part of a persons overall estate planning, they should cost no more than $250 to $500.

Todd Cordell, vice president of Chicago Trust Co. in Hinsdale, said establishing an estate plan can cost $500 to $3,000 or more depending on its complexity. It should be little to no extra cost to establish a pet trust as part of that estate plan, he said. If provisions of the pet trust need to be changed, amendments to the trust could cost $300 to $1,000 depending on the complexity.

It is recommended that the estate plan provide instructions on how to fund the pet trust. The amount will depend on such factors as the pets age, cost of routine care, food and toys, Cordell said.

In Illinois, a judge has the authority to reduce the amount of money used to fund a pet trust if the amount is deemed to be excessive.

Because pets are considered property, they must be bequeathed to the trust.

For most pet lovers, animals are part of the family. In fact, the Friedman family of Deerfield calls their high-maintenance 12-year-old Portuguese water dog, Payton, their oldest child.

The Friedmans estate plan, handled by Lindsey Paige Markus of Chicago law firm Chuhak amp; Tecson, calls for Payton to be adopted in the event of the premature deaths of dad, Brad, 38, an executive vice president for Chicago-based real estate development firm Urban R2, and mom, Jolie, 35, who works at Northbrook childrens clothing store Peek A Boutique.

While the couples two human children, boys ages 8 and 6, would be cared for by Brads brother and his wife, the Friedmans decided they are not suitable parents for a dog.
